Load both sides of the hemacytometer with the cell suspension - covering both counting grids. Count the number of cells in 5 out of 9 squares from each counting grid (count a total of 10 squares). Divide the total number of cells counted from 10 squares by 10 to get the average number of cells per square. Multiply the average count per square by 104 to get the number of cells per ml of the original cell suspension. Determine the grand total number of cells available for subculturing, multiply the number of cells per ml by the total number of mls of cell suspension.
